Departs from the terminus at the corner of Cotham and Burke Roads and runs via Burke Road, Whitehorse Road, Creswick Street, Kitchener Street, Wolseley Crescent, Gordon Street, Burke Road, McArthur Road, Lower Heidelberg Road, Marshall Street and Norman Street to Ivanhoe Railway Station. It then runs via Norman Street, Seddon Street, Upper Heidelberg Road, St. Elmo Road, Banksia Street, Edwin Street, Altona Street, Waterdale Road, Southern Road and Waiora Road to the terminus at the corner of Waiora Road and Wattle Avenue. The route then extends via Wattle Avenue, Main Drive and Plenty Road to the LaTrobe University Bus Interchange located at the corner of Plenty Road and Kingsbury Drive and then becomes Route 548 to Kew - see below.

ROUTE 548 from LaTrobe University Bus Interchange to Cotham Road, Kew:

Departs from the terminus at LaTrobe University Bus Interchange and runs via Science Drive, Waterdale Road, Kingsbury Drive, Waiora Road and then via the reverse of the route described above for Route 547 to the corner of Gordon Street and Burke Road and then continues along Burke Road to the terminus at the corner of Cotham and Burke Roads.

Route 548 from Cotham Road, Kew to LaTrobe University Bus Interchange:

Departs from the terminus at the corner of Cotham and Burke Roads and runs via the route described above for Route 547 to the corner of Waiora Road and Ruthven Street. It then runs via Kingsbury Drive, Waterdale Road and Science Drive to the terminus at the LaTrobe University Bus Interchange. The route then extends via Kingsbury Drive, Plenty Road, Main Drive and Wattle Avenue to the corner of Waiora Road and Wattle Avenue and then becomes Route 547 to Kew - see below.

Route 547 from Mont Park to Cotham Road, Kew:

Departs from the terminus at the corner of Waiora Road and Wattle Avenue and runs via the reverse of the route described above for Route 547 to the corner of Gordon Street and Burke Road then continues along Burke Road to the terminus at the corner of Cotham and Burke Roads.

• Times marked in "W" indicate trips that will be operated by ultra low floor, wheelchair accessible buses.
• Times marked "G" do not operate via LaTrobe University Bus Interchange but extend to the intersection of Grange Boulevard and Main Drive.
• Times marked "L" terminate at LaTrobe University Bus Interchange.
• Times marked "B" indicate buses depart from the corner of Burke and Belmore Roads.
• Times marked "R" indicate buses which depart from the corner of Ruthven Street and Waiora Road. Times shown indicate departure time from that point.
• Times marked "S" indicate buses which operate on school days only.
• Saturday service only operates on Route 547 to the intersection of Grange Boulevard and Main Drive via Mont Park.
• There is no Sunday service on Route 547.
• There is no weekend service on Route 548.
• Christmas holiday weekday timetable operates for a period of six weeks from mid December to Australia Day. Copies of Christmas timetables are available from the driver in early December.
• Easter Saturday operates to normal Saturday service.
• There is no service on Public Holidays except Labour Day and Melbourne Cup Day. Special timetables apply on these days and are obtainable from the driver one week prior to the holiday.
